It means I don't have to constantly check and see if Derek Sivers or Tynan has published a new post-it just pops up in my feeds. I follow a few dozen tech sites, but it's also really great for following blogs that only publish a few times a year. For more details on our process, read the full rundown of how we select apps to feature on the Zapier blog.Īs a tech writer, I rely on my RSS app to keep me up to date on what's going on. We're never paid for placement in our articles from any app or for links to any site-we value the trust readers put in us to offer authentic evaluations of the categories and apps we review. We spend dozens of hours researching and testing apps, using each app as it's intended to be used and evaluating it against the criteria we set for the category. My picks for the best RSS readers are far nicer than Reader ever was.Īll of our best apps roundups are written by humans who've spent much of their careers using, testing, and writing about software. The world of RSS apps has moved on and, a decade later, is actually in a much better place than it likely would have been if Google had remained at the top. While it's still traditional to bemoan the death of Google Reader all the way back in 2013 in any article about RSS, I'll skip the eulogy. You just open your RSS app and get reading, with every article and blog post presented in reverse chronological order. Still, it remains the absolute best way to combine stuff from loads of different places into one central app, where you can read it without having to click around a bunch of sites or scroll through your social feeds. Although pretty much every podcast app relies on RSS, it isn't as publicly popular as it used to be. RSS (it stands for Really Simple Syndication) has been around since the '90s, and it's a way for sites to publish a feed of all their content in a way that can be easily parsed and aggregated by RSS apps. ![]() ![]() The process couldn’t have been easier, and it’s worth noting that they offer free shipping. We went for the simplest option they offer: choose a frame, upload a photo, pay, and wait for it to arrive. They send four designer-chosen frame samples to your home so you can pick in person. The former is free and similar to a try-on glasses service. They also offer design services to help you choose a frame or hang a gallery wall. Could the packaging possibly work for a gift? As our framed photographs arrived we judged each one on four criteria giving each a numerical rating from 1 to 5. Things that stood out were wide arrays of frames and mats to choose from, free basic retouching, PDF proofs, image adjustments, and choice of hardware. Below, the results.ĭuring the ordering process we took notes on each retailer’s user experience. We also opted for frames on the small side to accommodate all kinds of placement and because a giant framed photo might be more of a burden than a gift. To accurately test for print quality across retailers, we chose to only use images taken with an iPhone 7. To find the best online photo print services (ahem, Mother’s Day gifts) we tested a handful of online-framing services, focusing on sites that let you upload a photo, choose a frame and mat, and purchase straight from a website or an app. There are tons of online-framing services that will take photos straight from your phone or Instagram feed and turn them into framed works of art. (Right now, I have just over 15,300 photos stored on my phone.) In the past, it was unlikely that any of those photos would make the journey from pixels to actual ink on paper, but in 2019 printing and framing photographs is actually easier than you think. ![]() With smartphones, each of us is walking around with tens of thousands of photographs of our friends, family, and pets every day. ![]() ![]() ![]() This poignant scene recalls earlier statements of Jesus: the promise that “the hour is coming when all who are in their graves will hear his voice and will come out” (5:28-29a) and that the shepherd “calls his own sheep by name and leads them out” (10:3). “Unbind him, and let him go,” Jesus commands (11:44). Obediently, Lazarus comes out, probably stumbling with his grave clothes still wrapped around his face and body. After repeating his promise that this would all lead to the glory of God, and praying out loud for the benefit of the crowd, Jesus cries, “Lazarus, come out!” (11:43). Jesus is undaunted by the stench of death. Does Jesus really understand about death? “Lord, already there is a stench because he has been dead four days” (11:39). Even though she had boldly professed to Jesus when he arrived, “Even now I know that God will give you whatever you ask of him” (11:22), she is not so confident now that Jesus knows what he is doing. When they come to the tomb, he says, “Take away the stone” (11:39). Of course, Jesus does more than share in the pain and sorrow of his friends. But it is clear that he is completely with them in their pain and loss, deeply moved and grieved. He doesn’t explain to Mary and Martha and all those grieving why he didn’t come sooner and prevent Lazarus from dying. Jesus does not answer all the questions that we might wish he would answer. John does not recount a verbal response by Jesus to Mary, but tells us that “when Jesus saw Mary weeping, and the Jews who came with her also weeping, he was greatly disturbed in spirit and deeply moved” (11:33).Then, after asking where Lazarus is laid, Jesus himself begins to weep (11:34-35). Jesus pulls the hope of the future resurrection into the present, promising abundant, eternal life that begins here and now. Those who believe in me, even though they die, will live, and everyone who lives and believes in me will never die” (11:25-26). After Martha confirms her belief in the resurrection on the last day, Jesus responds with another promise: “I am the resurrection and the life. To Martha, he responds with a promise: “Your brother will rise again” (11:23). It is noteworthy that Jesus does not rebuke Martha or Mary or their friends for what they say. Some of the neighbors gathered also ask among themselves, “Could not he who opened the eyes of the blind man have kept this man from dying?” (11:37).Īren’t these exactly the kinds of questions we ask, or would like to ask, when tragedy strikes? Where were you, Lord? How could you have let this happen? Couldn’t you have prevented all this horrible pain and heartache? Where were you, Jesus? Why did you take so long getting here? I thought you loved my brother. ![]() Implied in this statement are some pointed questions, perhaps even accusations. We can understand the anguished cries of Martha and Mary to Jesus, who greet him separately but with the same words: “Lord, if you had been here, my brother would not have died” (11:21, 32). Jesus expresses confidence that Lazarus’ illness will not lead to death, but rather to the glory of God (11:4), yet by the time Jesus finally gets to Bethany, Lazarus is already dead and buried. “Though Jesus loved Martha and her sister and Lazarus, after having heard that Lazarus was ill, he stayed two days longer in the place where he was” (11:5-6). ![]() Jesus’ response to their situation seems surprisingly nonchalant. Lazarus has fallen gravely ill, and his sisters Mary and Martha send for Jesus, who is “across the Jordan” (10:40 11:1-3). Mary, Martha, and Lazarus, three adult siblings from Bethany, are friends and followers of Jesus. Situated just after the “good shepherd” discourse and just before Jesus’ anointing and final entry into Jerusalem, the story is pivotal to the plot of John’s gospel and lifts up central theological themes of the Gospel as well. This story is rich in literary and theological themes interwoven with what has gone before in John’s gospel and with what is to come. ![]() ![]() ![]() As with all steps, optimization, 9 in this case choosing the “right” antibody and determining the optimal concentration, is key to a good blot.įigure 3: Examples of colorimetric, chemiluminescent, radioactive and fluorescent detection using an indirect method.īoth colorimetric and chemiluminescent detection require conjugation of an enzyme to the detection antibody and are considered very sensitive techniques. Then, a secondary antibody is used that binds the primary antibody and is detectable. Here, a primary antibody is used to probe the membrane and bind any target protein present. ![]() While it is possible to use direct detection (a single antibody that recognizes the target and is detectable) for a western blot, more often an indirect method is applied (Figure 2). Commonly used buffers include tris-buffered saline (TBS) and phosphate-buffered saline (PBS), often with the inclusion of Tween 20 (TBST and PBST). Ensure the membrane is well covered with an appropriate buffer and apply gentle agitation to wash the membrane evenly without damaging it. However, over washing can diminish the target signal so it is important to optimize the number and duration of wash steps. Insufficient or uneven washing can lead to poor quality/patchy blots and high background. This is achieved by incubating the membrane with a proteinaceous liquid such as milk or serum.įollowing blocking, it is important to wash the membrane between each step to remove excess or unbound reagents. Transfer efficiency can be assessed prior to probing using a removable stain such as Ponceau S.ĭue to the high affinity of blotting membranes for proteins, after transfer it is important to block any remaining binding sites to prevent subsequent non-specific binding of the assay detection antibodies. Dry transfer is efficient and quick but offers less flexibility than the other methods. Semi-dry transfer is quicker and still offers flexibility but is less efficient than wet transfer for large proteins. Wet transfer is efficient and offers flexible buffering but is time-consuming. Within electroblotting, there are also multiple strategies for transfer, known as wet, semi-dry and dry transfer. Proteins from the gel are carried across and attach to the membrane tightly. Here, the protein gel is sandwiched against the transfer membrane and an electrical current is applied. A number of techniques can be used for transfer, including capillary transfer, diffusion transfer and vacuum blotting, but by far the most common due to its speed and efficiency is electroblotting 8 (also called electroelution or electrophoretic transfer). Proteins must be transferred from the protein gel to an appropriate membrane (typically nitrocellulose or polyvinylidene difluoride (PVDF)) to facilitate antibody probing. However, this type of control can be problematic when comparing models in which “control” proteins are differently expressed, such as degeneration models. Detecting expression of a ubiquitous protein that should be even between all of your samples, such as actin in whole cell and cytoplasmic samples, can also be used as a loading control and helps to ensure consistent transfer of protein samples to the membrane. Running a duplicate protein gel and developing with Coomassie stain 5 can help to remove this uncertainty as it will show the amount of total protein 6 in each sample lane and can reveal any loading inconsistencies. This could mean that there is twice as much of the target protein in that sample, or it could mean that more sample or a more concentrated sample has been loaded in one lane than the other. For example, when assessing a blot, the band from one sample may appear twice as bright as another sample. It is essential, especially when trying to compare protein expression between different samples, to know how much sample has been loaded as this may not be apparent from the blot alone. It is also important to load appropriate control samples and size marker ladders to enable interpretation of the final blot. Solids will impair the running of the gel and it is likely your protein of interest will remain in the stacking gel. If your protein of interest is in the insoluble fraction (e.g., cell membrane-bound proteins) investigate pretreatment methods to liberate and solubilize it first. For a clean image, samples are centrifuged to remove any solids, in order to load only the soluble fraction. The specific separation method chosen will depend on the aim of the analysis. This is typically achieved by protein electrophoresis, such as sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS-PAGE) or native PAGE, which separates proteins based on their molecular weight or charge. Figure 1: Overview of a western blot protocol.īefore a western blot can be performed, the proteins in the sample must be separated. ![]() ![]() ![]() The Constitution empowers Congress "To promote the progress of science and useful arts by securing for limited times to authors and inventors the exclusive right to their respective writings and discoveries" in Article I, Section 8, Clause 8. If granted, he would have exclusive rights to his invention for 14 years based on the Patent Act of 1790, and he could hope to reap a handsome profit from it. Whitney knew that if he could invent such a machine, he could apply to the federal government for a patent. ![]() Her financial support would be critical to his success. Whitney was encouraged to find a solution to this problem by his employer, plantation owner Catherine Greene. The one variety that grew inland had sticky green seeds that were time consuming to pick out of the fluffy white cotton bolls. Long-staple cotton, which was easy to separate from its seeds, could be grown only along the coast. There Whitney quickly learned that Southern plantation owners were looking for a way to make cotton growing profitable at a time when tobacco was declining in profit due to over-supply and soil exhaustion. Reluctantly, he left his native Massachusetts to assume the position of private tutor on a plantation in Georgia. But, like many college graduates, he had debts to repay first and needed a job. Whitney filed this petition with Congress because his patent was set to expire.Įli Whitney and the Need for an InventionĪ recent graduate of Yale, Eli Whitney had given some thought to becoming a lawyer. Though he pursued a solution to this patent infringement in the courts, he was not always successful. Due to a loophole in the 1793 patent law, competitors were able to make cotton gins without his permission. Robert Herjavec asks to come up and try the Sweep Easy for himself, being that he was a stay at home dad for 3 years and is able to relate to these floor cleaning struggles. He wanted to create something that would make the lives of parents just a little bit easier. He has 3 children of his own and it’s definitely a bit of a challenge when it comes to keeping up with the kids while also being in charge of household tasks. In spite of that though, Shane believes wholeheartedly that this product will be a success on the market because he knows exactly what it’s like to have your hands full when you’re a stay at home parent. All he has currently are prototypes that he has made himself. He has yet to make any sales, but that is only because he doesn’t have an official product yet. What makes matters even better for the Sharks is that Shane currently has a patent pending status for this new broom design. Then, once the mess has been scraped up, you just retract the scraper and sweep up the rest of the mess. ![]() By pressing the top handle of the Sweep Easy broom, a scraper emerges from the broom and allows you to scrub off the dried mess on the floor without bending over. The question that Shane poses is what about the sticky messes that get stuck to your floors? No one ever enjoys getting on their hands and knees to scrub a messy floor, and that is why he reinvented the household broom. He then moves on to explain that, even though your typical broom has been a trusted cleaning appliance for decades, it is only good for merely sweeping things about the floor. Shane enters the Shark Tank seeking a $40k investment for a 25% stake in his Sweep Easy business. All he needed was someone with a little more experience in the business realm to help him get his foot in the door, and what better place to do that than the Shark Tank? However, will any of the Sharks be interested in investing into a company that has yet to make any sales? However, Shane didn’t let this dishearten him because he knew that parents everywhere could appreciate the Sweep Easy. A good portion of them said that they would definitely be interested in purchasing a few of them, but he has yet to receive any orders. ![]() He took Sweep Easy to movie theaters, super markets, and even schools. Once he was satisfied with the design, he went around to various public places to see if any business owners would bite. Shane turned his garage into a workshop and there he began to work on creating a prototype for the Sweep Easy.
